The Japanese slides you mention is probably "Database design practices for Social Games". This was more database design oriented talk rather than focusing on MySQ onlyL. I have not presented in English yet. If there are any good places to present, I'll do it:)

I used my own simple multi-threaded, O_DIRECT file i/o program. You can do almost same thing with fio (http://freshmeat.net/projects/fio/).

You reference ext* (ext3 and ext2 in particular) up against XFS. With multiple threads writing to the same file (using O_DIRECT to open), can we expect the same -- is ext4 included in "ext*" -- issues, with the numerous allocation changes made in ext4? Is XFS preferred over ext4?
Have you tried to use hugetlbfs [1]? I't would be nice to see some benchmarks on your environment.
